Born in Philadelphia, Clark Johnson moved with his parents and older sister Taborah to Toronto. He went on to attend Concordia University in Montreal, Quebec, then returned to Toronto where he made his television debut with a guest appearance on the long-running CTV series The Littlest Hobo in 1981. He made guest appearances on several more Canadian series including Alfred Hitchcock Presents, Katts and Dog and My Secret Identity, as well as small roles in American TV movies shot in Canada such as CBS-TV's Mazes and Monsters (1982) and Seduced (1985), and ABC-TV's Starcrossed (1985) before landing a regular role on the series Hot Shots (1986). The show only lasted one season, but Johnson was soon chosen to be a regular character on the hit series Night Heat as Detective Dave Jefferson from 1986 to 1988.

In the late 1980s, he began breaking through into major Hollywood features such as Adventures in Babysitting (1987), Colors (1987) starring Sean Penn, Iron Eagle II (1988) starring Louis Gossett Jr. and Renegades (1989) starring Kiefer Sutherland. In 1989, he became a regular on CTV's hit series E.N.G.

In 1991 Johnson made his directing debut with the Family Channel TV series African Skies, starring Catherine Bach and Robert Mitchum. In 1993 he not only landed a regular role on the new NBC drama Homicide: Life on the Street, but also directed several episodes. While continuing to act in films and television, Johnson began to get more and more directing jobs on both sides of the border, including episodes of ABC-TV's police drama NYPD Blue, CTV's Nikita, and NBC-TV's Law & Order: Special Victims Unit, The West Wing and Third Watch.

Johnson made his feature film directorial debut with S.W.A.T. (2003), starring Samuel L. Jackson and Colin Farrell. Johnson's younger sister Molly is a well-known Canadian singer, and his older sister Taborah, also well-known in Canada, has acted alongside Clark in the movie Blood Brothers (1993) and was directed by him in an episode of Homicide: Life on the Streets.
